Abstract : The aim of this project is to study the effects of the MTBE, which widely used in Saudi Arabia and other countries, it is added to unleaded gasoline to boost octane and enhance combustion. Recent studies proved that methyl tertiary-butyl ether (MTBE) has serious hazards to the environmet and human health. The present study thus aims to study the effects of chronic administration of MTBE on locomotor activity behavouir, and physiological functions of the liver and kidneys and study its effect on some neurotransmitters (dopamine, norepinephrine and serotonin) in different brain areas in albino mice. It was found during this study that the above MTBE has inhibitory effect on the Locomotors behaviour. There was significant decrease in the Latency (second) to first movement and Number of wall rears. The biochemical analysis of the blood in second and thread groups showed significant increase in the level of Liver enzyme such as (SGPT), (SGOT), (Alk phosphates) and Bilirubin. Also when albino mice exposed to inhalate gasoline vapor contaminated with MTBE both in gas station and the lab (6 h. a day, 5 days per week for 3 months) caused significant decrease in the content of norepinephrine, dopamine and serotonin in different brain areas of albino mice .The content of norepinephrine decreased significantly in all tested brain areas except in striatum and cerebral cortex. The content of dopamine decreased significantly in all tested brain areas except in cerebral cortex were it caused non-significant change. On the other hand the content of serotonin decreased significantly in striatum, hypothalamus, midbrain and hippocampus only. The decrease in the content of norepinephrine, dopamine and serotonin may be due to the neurotoxic effect of MTBE which affect in GABA A receptors which play an important role in release of these neurotransmitters .
